The Canadian Agency for Drugs and Technologies in Health (CADTH) Rapid Response Service has produced a review of the evidence on the safety of leflunomide and methotrexate as combination therapy for rheumatoid arthritis. The evidence was limited and definitive conclusions were not possible. One RCT in a systematic review on DMARDs and one non-randomised study on DMARDs suggested an increased risk of abnormal liver function for treatment with the combination.
